The pancreas is the answer to almost every mixed-gland question. Its exocrine part secretes digestive enzymes into the small intestine through a duct, while its endocrine part — the islets of Langerhans — releases insulin and glucagon into the blood. The gonads work the same way, producing gametes and hormones.

The simplest way to hold the distinction is by the duct. Duct present means exocrine; no duct means endocrine. That single test answers the classification questions in both directions.

Liver against thyroid is the pair that catches candidates. The liver is the largest gland overall but it is not an endocrine gland; the thyroid is the largest gland that is endocrine. Papers place both in the options and rely on the qualifier being skimmed.

The pituitary is called the master gland because its hormones control several other endocrine glands. Note that it is itself regulated by the hypothalamus, which is why some questions describe the hypothalamus as the true controller.

3. What enzymes are

Enzymes are biological catalysts — they speed up reactions in the body without being consumed. Almost all are proteins, and the properties below are asked as their own questions.

  • Specificity. Each enzyme acts on a particular substrate, which is described by the lock-and-key model.
  • Optimum temperature. Human enzymes work best around 37 degrees Celsius; high temperature denatures them.
  • Optimum pH. Each has its own — pepsin needs an acidic medium, trypsin an alkaline one.
  • Not consumed in the reaction, so a small quantity works repeatedly.
  • Denaturation by heat, extreme pH or heavy metals destroys activity permanently.

Two exceptions are worth knowing. Ribozymes are RNA molecules that act as enzymes, which is the standard answer when a question asks whether all enzymes are proteins. And bile is not an enzyme at all — it is a secretion that emulsifies fats into smaller droplets so that lipase can act on them. That is one of the most common trick questions in this topic.

Three points come up again and again. Digestion of starch begins in the mouth with salivary amylase, not in the stomach. Digestion of protein begins in the stomach with pepsin. And most digestion and almost all absorption happen in the small intestine, which is why the largest group of enzymes sits there.

Pepsin is secreted as inactive pepsinogen and is activated by the hydrochloric acid of the stomach; trypsin is secreted as inactive trypsinogen and is activated by enterokinase. Both are asked as activation questions, and the pairing of each inactive form with its activator is the exact form the question takes.

Hydrochloric acid itself is not an enzyme. It provides the acidic medium pepsin needs and kills bacteria in food, and it is secreted by the parietal cells of the stomach.

Revise the enzymes by location rather than alphabetically — mouth, stomach, pancreas, small intestine. Questions usually give the enzyme and ask where it is secreted or what it acts on, and grouping by organ means one recall pulls its neighbours with it. The small intestine holds the largest group, which is itself worth noticing.

Then fix the four traps separately, because that is where marks are actually lost. Bile is not an enzyme. Liver is the largest gland but thyroid is the largest endocrine gland. Pancreas is mixed. Pepsinogen is activated by hydrochloric acid while trypsinogen is activated by enterokinase.
